I see Mito Labs has both Tirzepatide and Retatrutide now. From what I understand:

  • Tirzepatide = dual agonist (GIP + GLP-1)
  • Retatrutide = triple agonist (GIP + GLP-1 + Glucagon)

So Retatrutide is the "newer generation" version? The triple mechanism apparently targets fat loss more aggressively but I'm worried it might be too strong or have worse side effects.

Has anyone tried both? Or switched from Tirzepatide to Retatrutide? What was the difference like?

Also curious about dosing — do you start lower with Retatrutide since it's more potent?

I've been on Tirzepatide for 3 months (see my results post) and I'm considering switching to Retatrutide for my next cycle. From the research, the glucagon receptor agonism adds thermogenesis — literally burning more calories — on top of the appetite suppression. But start even lower than Tirzepatide. 2mg/week for the first month minimum.

Retatrutide is the next generation, yes. The triple mechanism is more aggressive for fat loss but also means more potential GI side effects initially. My recommendation: if you haven't used GLP-1 agonists before, start with Tirzepatide. If you've done well on Tirzepatide and want to step up, Retatrutide is the logical next move.

The glucagon component is what makes Retatrutide unique — it actively promotes lipolysis rather than just reducing appetite. Early data shows slightly better fat loss than Tirzepatide at comparable doses. But as Dan said, start very conservatively.

I had a client switch from Tirzepatide to Retatrutide. Lost an additional 4kg in the first month after plateauing on Tirz. The transition was smooth since her body was already adapted to GLP-1 agonism. Worth considering if you've hit a plateau.

Hi Lisa,

Great question! Both Tirzepatide and Retatrutide have shown promising results in weight management, but they do have some differences that might influence your decision. You're spot on about Tirzepatide being a dual agonist and Retatrutide a triple agonist, which essentially means Retatrutide not only targets appetite suppression and insulin sensitivity but also adds a thermogenic effect through glucagon activation. This can potentially lead to more pronounced fat loss, as some in the community have noted.

When it comes to dosing, it’s wise to start lower with Retatrutide, especially if you're transitioning from Tirzepatide. The additional glucagon pathway can make it feel a bit more potent, and as others have mentioned, there's a possibility of increased GI side effects at first. Starting at around 2mg/week sounds like a reasonable approach until you see how your body responds.

If you've been doing well on Tirzepatide and are considering stepping up your weight management game, Retatrutide could be an exciting next step. Just keep track of how your body adjusts and consult with a healthcare professional if you have any concerns about the transition. Let us know how it goes if you decide to make the switch!
